When can I leave an Assistant behind, and can I leave more than one?
The designer ruled you must leave the Assistant as you move, and the rulebook describes the option at that moment.

Only at the moment you move from one Location to another, and only on the Location you are leaving, before you take the action at the new one. You can't leave one at the end of your turn, at a Location you left earlier, or with an Assistant you just hired. Over several turns you can have one left behind at each of several Locations, but never two on one space.
