Do I have to defeat the enemies on a Vortex delve tile before I enter it?

Only if you want its skyshards. The Vortex card's skyshards disappear if you enter the tile while enemies are on it, so clear them first by ranged attacks or by drawing them off. You can also stop your movement short of the tile.
