Must I select a Blessing at Gear up for every test?

No. Using a Blessing is optional like other gear, but to use it on a test you must select it at Gear up before drawing. The lock icon only prevents discarding or giving it away. Conditional options such as "if you have a Blessing" are satisfied by having it in your purse, and the same goes for Weaknesses unless a Weakness has a mandatory effect.
