How does a glyph work: is it an action, how often do I pay, and does it depend on where my hero stands?

A glyph is not an action. It only adds a 1 spirit cost to the matching action while it faces your home kingdom, paid every time you take that action on your turn. It follows the player whose home kingdom it faces, not the hero's current kingdom. If you can't pay, you simply can't take that action, with no corruption. Any other requirement for the action (like skulls to Cleanse) still applies.
