Can spare colonists on my windrose staff a newly built building or plantation?
The rulebook states this directly, and forum players agree.

No. They are placed only in the next Mayor phase; colonists can only be placed on tiles during the Mayor phase. A new tile gets a colonist outside the Mayor phase only through the Hospice, University or Guest House.
