Yes to both. When a survivor fills an Age box you roll on the Age chart at once and apply the result; weapon proficiency has no chart and simply grants specialization or mastery. A milestone can be gained once per lifetime, so losing Hunt XP does not remove the bonus and refilling the box doesn't repeat it.
