Do Chest Tiles respond to Criminal icons or count in end-game Manuscripts?

No to both. Criminals are not wild for Chest Tiles unless the tile is keyed to a Criminal, and Chest icons do not count for Manuscript scoring since cards never move during scoring. An x1 chest counts each icon only once.
