Does the Priestess of Mysticism have to be at the Dark Alley to use her power?

No. With her original Fate Weaving ability the player only needs two Dark Alley AP; the Priestess need not be the one placed there. Dahlgaard's Academy recommends replacing this ability with Deja Vu (pay a Shard to keep a placed Special Assignment card); with that version and Enhance Trick, you cannot stack extra shards on the same trick, since the card adds +1 only once.
