For the 9-square column and 12-square row lessons (221, 222), must the whole run be filled with one colour?

Yes. The run of 9 or 12 squares must be all cats of the chosen colour, with no gaps and no treasure. The row is not the full length of the boat; it is any straight run of 12 squares in a line. Other cats elsewhere in that row do not invalidate it.
