No — R5-K6 lets you roll a defense die after spending your lock and, on an evade result, re-acquire a lock on the same ship, but the card forbids spending that new lock during the current attack. It is there for your next attack. A lock R5-K6 reacquires is removed like any other if its target is destroyed.
