Do the two sides of the reference card change any rules by player count?
No. Barrel costs, Rum-runner strength and the building hierarchy are the same on both sides. The only difference between the 1-2 player side and the 3-4 player side is the Zone Control payouts. The card comes with the upgrade pack, which is why the base rulebook does not mention it.
