Do I lose my Associate if all that Mobster's Speakeasies are taken over?

No. You keep the Associate and its benefits, and it can still help you defend or take over. A Mobster tile also stays on the board after losing all Speakeasies, so no one can remove another player's Associate by taking over buildings.
