Can I use the New Starting Worlds promo with the basic start worlds, and should I in a first game?

Yes to the first; no to the second. Shuffle the red and blue start worlds separately and deal one of each to every player, as the sidebar on page 8 of the expansion rules describes. The designer advises new players to skip this in their first games because choosing a start world is an extra decision.
