At the start of your turn to take cards, after drawing has ended, if five or more cards are in the harbor. It does not trigger when the fifth card is revealed, and you get the coins even if you then take nothing. Ships are not the only cards that count, but Tax Increases, Missions and Expeditions never enter the harbor.
