Are both Mason cards always in the game, and are there always two Masons?
The rulebook says to always use both Masons and that a missing partner is in the center.

Both Mason cards always go into the game together when Masons are used, but either may be in the center, so only one player (or none) may actually be a Mason. A lone Mason learns that the other Mason card is in the center.
