No. A discovered Weakness works once your Knowledge reaches its required level (3, 5 or 8), and Item requirements count your Knowledge whether or not the token is active. Activation matters for the Archive action, which gives 2 extra Knowledge, and some Objectives. Gaining Knowledge further does not require activation.
