Nemesis Survivors can: the first Weakness starts revealed, and a Survivor, Lab Rat or other Nemesis Survivor who starts at Knowledge 3 can exploit it at once. Facility Workers cannot use it until some Character has researched an Object. With classic Intruders the first Weakness starts undiscovered instead.
