Can I combine a village action, a quest or an assistant action on the same stop?
The rulebook allows one main action per turn; the designer lists the four main actions.

No. Each turn you perform one main action: a city action, village actions, fulfilling a quest at your location, or one assistant action. A village with a quest tile therefore offers the quest or the village actions, not both, and an assistant action there replaces village actions as well. Effects that explicitly grant extra village actions are the only exception.
