Are cards on the 5th tower coloured for colour-specific effects?

Yes while on the tower, no once you take them. Cards on the 5th tower are colourless, so an effect aimed at one colour (such as taking a blue card from the towers at a discount) does not apply to them. Once you take the card it regains its colour, so effects that trigger when you take a blue, green, yellow or purple card do apply. Effects not tied to colour (take a card from the towers) still work on the 5th tower.
