Does completing a Mandatory Quest use up my one quest for the turn, and must I do it first?
The rulebook says you cannot complete other quests until the Mandatory Quest is done.

Yes to both. A Mandatory Quest is a quest, so it is your single quest for that turn, and you cannot complete any other quest until it is done. After completing it, it goes to the Intrigue discard pile, never to your completed quests, so it does not count toward Lord bonuses.
