Can I put more than one agent on Cliffwatch Inn or Waterdeep Harbor, and when is the quest row refilled?
The rulebook states the immediate replacement and the three spaces.

Yes, if a space is open: each has three action spaces and you may use more than one over several turns, but only one agent per turn. A face-up quest you take is replaced immediately from the deck, not at round end, and the Inn always holds four face-up quests.
