Do I need a transport tile with an upgrade symbol to upgrade, even when only skills are needed?
The Transport and upgrading section ties upgrades to those tiles' upgrade icons.

Yes. Upgrading happens only by placing a worker on a transport or upgrade tile (home tile, Farrier, Stable, Wainwright), and its upgrade icons set how many tiles you may upgrade. You do not activate the tile being upgraded.
