The rulebook ties the Brenn and Advantage card handout to the Assembly; chieftain status itself is simply who has the most clans.

Yes, immediately: whoever has the most clans at that moment is chieftain, which matters for Withdraw and card effects. The Brenn changes only at the Assembly and Advantage cards are handed out only then, though you keep one you hold.
