Do the two tiles placed on the board during setup use their effects?
One forum reply confirms it; the rulebook only says effects follow playing a tile on your turn.

No — the two tiles drawn onto the central spaces during setup are simply placed and trigger nothing. Effects only come from tiles you play from your hand on your turns.
