Yes, through the Swap Heroes option in the app settings, and you can use it repeatedly. The publisher says this usually restarts the current Act, since checkpoints sit between Acts, so time it near an Act boundary. The printed rulebook says swapping heroes needs no Act restart, but the publisher's description of the app is the one to expect. On a restart the app keeps your story choices from the start of the Act. You can also keep up to four campaigns in separate save slots, and the app adapts if you drop from four heroes to three.
