Do I get the Neighbourhood bonus again when I place another unit next to the same opponent unit?

Yes. The bonus fires whenever a unit you place is adjacent to an opponent's unit, so a later placement beside the same unit earns it again; it is not once per opponent or per settlement. You need a free merchant and the money each time.
