Must a new tile line up with the existing grid?

Yes. A new tile must share a whole edge with an existing tile, except along the long sides of the starting tile, where it lines up with either half. Tiles are never stacked or placed off-grid, and the purple (safe) tiles can go anywhere your routes lead.
