Can I play a card that says 'when a hero attempts a challenge' after the runes have been cast?

Yes. A challenge is not finished until its effects resolve, so cards worded 'as any hero attempts a challenge' can modify the attempt after the cast. Cards worded 'before any hero attempts' must come before the runes are cast.
