Is Ekolu's ability to clear all 3s after her third pattern mandatory, and does the pattern position matter?
It is mandatory, since the rules would say "may" otherwise. It triggers after your third pattern in the round, wherever that pattern was in your hand, and all 3s in sight go to the opponent automatically.
