Do I need at least one qualifying piece to score an Objective tile, for example second place in a 2-player game?

Per a 2021 publisher post, yes: you need at least one piece on the objective to score it, and the rule was changed from an earlier ruling that let second place take 10 VP with nothing built. No updated FAQ was ever published, and the digital versions (BGA, Steam) still follow the older no-build ruling, so tables disagree. A region or area with zero of your structures simply counts as 0.
