What do the setup tiles and X markers on the Duel map mean?
The cardboard setup tiles block building on their numbered spaces: the X means you cannot build there and they are not conduits to use. The two neutral dams are replaced by wooden neutral dams on the same spaces, and no unused player color is needed. The final printed rulebook says this, unlike the earlier online version.
