Can I raid a starport, and can I raid after destroying the defender's only building?

Yes to both. Starports are buildings, and you can raid them, as long as you still have an attacking ship once the hits are resolved. Raid keys are spent one at a time after the hits, even if the building was destroyed in that battle.
